Dregs Analysis Skill¶

An AI skill for the process in Isolating Organisms. You give it a bottle you want to harvest; it researches the beer, reads your dregs micrographs and writes an isolation plan that uses only what your kitchen-table set-up can do. Everything here is experimental home practice: read the disclaimer first.
What it does¶

How it plans:
- From a capability matrix. Every item and procedure is Available or Unavailable, and each procedure lists what it needs. The skill takes the standard (primary) route for each step when it can, and otherwise the best feasible fall-back, stating the cost in standard terms (reduced selectivity, composition bias, longer timeline, lower identification confidence and so on) and the purchase that would restore the primary route. Availability is checked when a plan is written; ask for a matrix review to re-check a plan after your kit changes.
- Plate before enriching, keep a mother culture, selectivity in the plate, one target per plate type, calibrated dilutions, controls for home-made media, hop pressure for beer bacteria.
- Safety policy. It never suggests prescription or veterinary medicines, or handling pure antibiotics or cycloheximide.
How to install and use it¶
- Download the zip.
- In Claude, install the skill by uploading the zip (Settings → Capabilities → Skills), or place the folder where your Claude set-up reads skills. Installed at account level, it is available in every project.
- Make it your own: copy
data/default-capability-matrix.md(the author's set-up), set Status and Held for your kit, keep the IDs, and give the skill the file or its path. Without one it plans with the bundled matrix and says so. The author's live copy is the Isolation Capability Matrix. - Start with "I'm opening a bottle of …", with label photos and micrographs (objective, eyepiece field number, and one full-field frame showing the field edge for size calibration).
- Plans, critiques and merge drafts are Markdown files. With the author's wiki repo connected
they go to
work/isolation-plans/<bottle-slug>/; otherwise to the session's outputs.
scripts/dregs_calc.py (Python 3 with NumPy and Pillow for calibrate) fits the field stop on a
micrograph for scale, converts a wet-mount field count to cells per mL, and gives the
undissociated fraction of hop acids or sorbic acid at a given pH.
